Comparison of amniotic membrane versus the induced membrane for bone regeneration in long bone segmental defects using calcium phosphate cement loaded with BMP-2

Thanks to its biological properties, the human amniotic membrane (HAM) combined with a bone substitute could be single-step surgical alternative two-step Masquelet induced (IM) technique for regeneration of critical defects. However, no study has directly compared these two membranes. We first designed 3D-printed scaffold using calcium phosphate cement (CPC). assessed suitability in vitro support marrow mesenchymal stromal cells (hBMSCs) attachment and osteodifferentiation. then performed rat femoral size defect compare IM approach HAM. Five conditions were compared. Group 1 was left empty. 2 received CPC loaded rh-BMP2 (CPC/BMP2). 3 4 CPC/BMP2 covered lyophilized or decellularized/lyophilized 5 underwent two- step procedure insertion polymethylmethacrylate (PMMA) spacer followed by, after weeks, replacement wrapped IM. Micro-CT histomorphometric analysis six weeks. Results showed that supported proliferation osteodifferentiation hBMSCs vitro. In vivo, very efficiently formation led satisfactory healing defect, single-step, without autograft need any covering. this study, there difference between single approach. results indicated none tested membranes further enhanced group.
